OmniProt™ implements a nanoparticle-based low-abundance protein enrichment strategy for plasma and serum samples, enhancing proteome coverage and improving the consistency of quantitative mass spectrometry analysis.

OmniProt™ workflow

OmniProt™ is well-suited for lab-scale studies and rapid method development. Its flexible workflow allows customization at each step, supporting precise experimental control and accelerated optimization without reliance on fixed protocols. A reference protocol is provided below. [1]

①Sample

Mammalian plasma/serum samples (human or mouse).

 

②Enrichment
Incubate samples with OmniProt™ beads at 30 ℃ for 30 min to form protein corona.

③Lysis
Remove excess proteins, then lyse, reduce and alkylate to prepare proteins for digestion.

④Digestion
Digest proteins into peptides using trypsin.

⑤Cleanup
Desalt and purify peptide mixtures before LC-MS analysis.

⑥MS Analysis
Run LC-MS/MS using TMT (30 fractions) or Astral DlA (60 samples/day).

OmniProt™ working principle

· Dynamic protein corona - powered selective protein enrichment:

OmniProt™ uses nanoparticles that form a dynamic protein corona when exposed to plasma or serum, creating a biological interface that enables selective enrichment of proteins across various abundance levels. [1]

By engineering a protein corona through nanoparticle-plasma interactions, OmniProt™ significantly enhances the identification of low- and medium-abundance proteins in human and mouse plasma/serum samples.
